Simultaneous determination of baicalein, chrysin and wogonin in four Iranian Scutellaria species by high performance liquid chromatography

ABSTRACT

Abstract Scutellaria is a diverse and widespread genus of the Lamiaceae family of herbaceous plants known commonly as skullcaps. It has been implicated for medicinal purposes for the treatment of various ailments ranging from cancers, nervous disorders, hepatitis, anxiety and jaundice. A sensitive, simple and reliable high performance liquid chromatography (HPLC) method was developed for simultaneous quantification of three major active ingredients, chrysin, wogonin and baicalein in S. bornmuelleri, S. orientalis, S. araxensis and S. multicaulis. HPLC analysis was performed using a C8 analytical column. Detection was carried out at 262 nm using a mobile phase of methanol–acetonitrile–aqueous phosphoric acid at 1 ml min−1 flow rate. The test range of chrysin, wogonin and baicalein concentrations was from 0.5–400 μg mL−1. The experimental results revealed that the amount of baicalein in the roots of examined plants was more than shoots. Wogonin was more in the roots of S. orientalis and S. bornmuelleri and shoots of S. bornmuelleri and S. araxensis. The highest amount of chrysin was detected in the roots and shoots of S. orientalis and S. bornmuelleri. Totally the amount of flavonoids in S. orientalis shoot and root was more than other species. The high percentage of recovery (94.33–111.11 %), low limit of detection (LOD) and limit of quantitation (LOQ) and high R-squared value (R2 > 0.999) confirm the suitability of the method for simultaneous quantification of baicalein, chrysin and wogonin in the four Scutellaria species.
