Model-informed drug development and discovery: an overview of current practices

ABSTRACT

Abstract Mathematical modeling and simulation (M&S) tools are routinely utilized in a variety of disciplines (e.g., engineering, economics, meteorology, behavioral sciences, aeronautics, and space travel) and have been responsible for expediting the development of many fields. As integration of mathematical modeling in life science course work has largely been under emphasized, such approaches have generally been implemented by professionals coming from engineering-based disciplines, and thus considered foreign to the field, which hindered the rapid integration of M&S in pharmaceuticals discovery and development. The slow adoption of M&S techniques in drug discovery and development has had a profound negative impact on the productivity and efficiency of pharmaceutical companies. The goal of this chapter is to build a conceptual framework that elucidates how different types of M&S activities are currently practiced in drug discovery and development. This chapter places a special emphasis on the application of model-informed drug development tools and approaches in the development of oncology compounds.
