ABSTRACT

Highlights • Automation led to the localization of FtsZ filaments and constriction mechanism.• CEMOVIS confirmed the presence of the mycoloic-acid outer membrane in Mycobacteria.• Cryo-FIB SEM illustrated the sporulation process and asymmetrical cell division.• Cryo-CLEM was used for the identification of a non-canonical T6SS.• Subtomogram averaging led to higher resolution by overcoming missing information reveals internal conformation changes and mechanisms.• Perspective of further coming advancements.
