Abstract This chapter describes structure and function of poliovirus RNA-dependent RNA polymerase (RdRP). It starts by providing the perspective for the need of antipolio drugs along with the various targets for the discovery of such drugs. A highlight is then presented on the importance RdRP of the virus in the development of drugs against it, followed by the discussion on the various inhibitors of the enzyme discovered till date and the challenges encountered in their designing and biological evaluation. The chapter concludes with a future perspective in the discovery of novel drug molecules against polio.
