Fluorescence detection and imaging in zebrafish and Arabidopsis thaliana based on Cys/Hcy breaking space effect

ABSTRACT

Abstract The discriminative detection of mercaptans has become a hot spot of research because of their obvious roles in biological systems. In our work, we constructed a complex, that perylene conjugated 2-chloropyridine by α, β-unsaturated ketones. The fluorescence of fluorophore perylene was quenched by the vertical linker. Cys/Hcy can break the space effect by nucleophilic addition to α, β-unsaturated ketones, which makes the system give a fluorescence emission. This probe was obtained via one-step reaction with good yield. Furthermore, the study showed the probe featured excellent selectivity and specificity for Cys/Hcy with a good reversibility. Moreover, this probe suggested a potential for imaging in vivo, which was confirmed by the successful imaging of the probe in HepG2 cells, zebrafish and Arabidopsis thaliana.
