Highlights • The four different reverse genetic systems developed for CoVs are reviewed and discussed.• These systems were based on the use of BACs, in vitro ligation and vaccinia virus as a vector.• CoV reverse genetics is an essential tool for the study of CoV replication, virus–host interaction and pathogenesis.• These reverse genetic systems allow the development of genetically defined vaccines and the analysis of antiviral drugs.
