Mammalian Liver Phosphoenolpyruvate Carboxykinase Activities

ABSTRACT

During an investigation of factors affecting phosphopyruvate formation from a-ketoglutarate by liver mitochondria (1, 2) great differences were found between species.’ Mitochondria from livers of guinea pigs, like those from rabbits (a), formed phosphoenolpyruvate in large amounts whereas similar preparations from rat livers formed relatively minor am0unts.r As has been suggested (2) and substantiated by us,l phosphoenolpyruvate carboxykinase (3) (GTP : oxalacetate carboxy-lyase (transphosphorylating); I.U.B., EC 4.1.1.32) could be involved in the over-all formation of phosphoenolpyruvate (P-enolpyruvate) starting with a-ketoglutarate and orthophosphate under aerobic conditions, in the presence of 2,4-dinitrophenol, by catalyzing Reaction 1.
