• Combining the GPD promoter with the Sed1 anchoring domain produced the highest β-glucosidase activity among the tested surface-display cassettes.
• The cell-surface-displayed β-glucosidase retained relatively high activity at pH 3.0 and at glucose concentrations below 2.5% (w/v), outperforming AR 2000 under those conditions.
• Varying ethanol concentrations did not produce a significant activity difference for the cell-surface-displayed β-glucosidase compared with AR 2000.
