There has been incremental evidence of miRNA binding sites not only being restricted to the 3′ UTR of mRNAs, but being consistently present in both coding regions and fewer examples in 5′ UTR regions. In this manuscript, the authors use a predictive algorithm to identify two putative miR-103a-3p in the 5′ UTR of the GPRC5A mRNA. The target mRNA is of interest in cancer progression, acting as either a tumor suppressor or an oncogene in different cancers. They clearly demonstrate that ectopic expression of this miRNA leads to a decrease in the levels of GPRC5A mRNA and protein in pancreatic cells. Interestingly, the authors show that miR-103a-3p targeting of the 5′ UTR but not the CDS of GPRC5A results in decreased levels of the corresponding protein.
